Temperature zones & envelope
GDP warehouses combine +2/+8 °C mainstream storage, −20 °C frozen (biologics/APIs) and optionally −70 °C ULT for mRNA. Envelope: 200–350 mm PIR, vapor barrier both sides, sub-floor heating below −5 °C zones.
Refrigeration architecture
Chilled water loop for +2/+8 °C (glycol back-up); NH₃ low-charge or NH₃/CO₂ cascade for −20 °C; discrete ULT cabinets for −70 °C. N+1 minimum; dual utility feeds and diesel N+1 with 72h autonomy.
Monitoring & 21 CFR Part 11
Wireless calibrated loggers (2 per 100 m² min.), central EMS with audit trail, alarm escalation, redundant server + off-site backup. Calibration NIST-traceable, annual re-cal.
Validation programme
URS defines scope and acceptance; DQ against URS; IQ verifies installation; OQ runs empty/loaded challenge; PQ runs seasonal mapping (min. summer + winter, 7-day). Deviation SOPs required for WHO PQ.
Procurement pitfalls
Avoid split GC + refrigeration + validation contracts without an integrator — hand-off gaps kill mapping timelines. Bundle EPC + validation partner + monitoring vendor from RFQ stage.
Preguntas frecuentes
What redundancy level does GDP require?
GDP does not prescribe N+1, but WHO PQ and most MoH audits expect zero-downtime architecture: N+1 chillers, dual utility feeds, backup generator with 72h fuel, and UPS on all monitoring.
Do I need a separate −20 °C room?
Yes for biologics, most vaccines and controlled substances. Envelope separation prevents cross-contamination and simplifies mapping; sharing walls with +2/+8 °C is allowed with proper thermal breaks.
How long does validation take?
Typical GDP+WHO PQ: 6–9 months from IQ start (empty mapping → loaded → seasonal). Add 2–3 months for CDSCO/MoH licensing in emerging markets.
